Automated RNA Isolation Service
The Core operates an Autogen 245T Nucleic Acid Purification System for walk-away isolation of RNA form solid tissues or cell pellets in 12 or 24 sample batch sizes. Unfortunately, this system cannot handle blood samples. The system uses a modified Trizol method yielding a wet ethanol pellet. In some cases, a DNA removal step may be needed for the final desired use. Since this system uses a Trizol based purification system without downstream column purification, it retains small RNAs including miRNA, tRNA and 5S rRNA and thus is suitable for small RNA isolation. Note that most commercial kits remove small RNAs. This is a rapid, low cost means to process large numbers of samples.

To Summarize:

1. We accept frozen whole tissue (on dry ice) or disrupted cell culture samples. Sorry we are not equipped to isolate RNA from blood.

2. Samples should be 5-100 mg (wet tissue), but vary by tissue type in the amount that is recommended per prep. Please consult with the lab if you want to submit smaller samples.

3. Typical yields are about 1 µg RNA per mg wet tissue, but vary by tissue.

4. Samples are processed in multiples of 12, maximum of 24 per batch.

5. For the best results, we strongly recommend that all samples be prepared during initial isolation using RNAlater (Ambion), if at all possible. We have found some tissues that will require the addition of RNAlater to prevent RNA degradation. Check with the lab for details.

6. We will supply you with RNA as a wet ethanol pellet. We can resuspend your RNA and check it on the BioAnalyzer for an additional fee.
